Target Information
Nerve growth factor (NGF) influences the survival and differentiation of various neuronal and nonneuronal cells. It binds to specific cell surface receptors with high and low affinity sites, where only the high affinity receptor mediates signal transduction. Cross-linking studies show that the high affinity receptor of rat PC12 cells forms a 158,000 Mr complex with NGF, while the low affinity receptor forms a 100,000 Mr complex.
Full-length cDNA clones for human and rat NGF receptor genes show over 90% homology. NGF stimulates nerve growth and regulates differentiation of sympathetic and sensory neurons. Mutations in the NGF gene are linked to hereditary sensory and autonomic neuropathy type 5 (HSAN5), and dysregulation of NGF expression is associated with allergic rhinitis.
