Target Information
MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily.
It acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway and diminishes T cell receptor (TCR) signaling and T cell proliferation by phosphorylating adaptor protein SLP-76.
HPK1 is expressed predominantly in hematopoietic cells, including early progenitor cells, and specifically activates JNK1, enhancing AP-1-mediated transcriptional activity.
Unlike PAK65, HPK1 does not bind to small GTPases Rac1 or Cdc42, indicating a distinct regulatory mechanism within the MAPK cascade.
