Product Specific Information

The antibody was affinity-purified from rabbit antiserum by affinity chromatography using epitope-specific immunogen. The purity is >95% (by SDS-PAGE).

Target Information

Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase that plays an essential role in the T cell activation pathway.
Calcineurin (also referred to as protein phosphatase 2B) is composed of two subunits:

  • Calcineurin A (CnA): 60 kDa catalytic subunit, homologous to protein phosphatases 1 and 2A
  • Calcineurin B (CnB): 19 kDa regulatory subunit containing four EF-hand calcium binding sites

Calcineurin-mediated dephosphorylation of the nuclear factor of activated T-cells (NF-AT) is essential for NF-AT activation, nuclear translocation, and early gene expression in T-cells.
Immunosuppressive drugs such as cyclosporin A and FK506, when bound to immunophilins (cyclophilin and FKBP respectively), inhibit NF-AT activation by calcineurin. This inhibition occurs as the immunosuppressant/immunophilin complex binds simultaneously to both CnA and CnB, blocking substrate access to the catalytic domain of CnA without inhibiting its phosphatase activity.
