Target Information

TRF1 is a negative regulator of telomere length maintenance. Two proteins, tankyrase and Tin2, bind to mammalian TRF1. Tankyrase localizes to telomeres by binding to TRF1 through its ankyrin repeats and exhibits PARP activity, functioning in ADP-ribosylation. ADP-ribosylation of TRF1 reduces its DNA-binding ability, suggesting regulation of telomere function via poly(ADP)-ribosylation.
TIN2 was identified as a TRF1-interacting partner from a yeast two-hybrid screening. It localizes to telomeres and is essential for telomere length regulation. Both TIN2 and TRF1 are constitutively expressed, acting together to counterbalance telomerase-mediated elongation. However, endogenous expression levels are low, making visualization by immunostaining challenging in some systems.
