Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties in vivo and has been effective in treating renal, heart, and liver allograft rejection. CD3 is expressed by thymocytes and all mature T cells, playing a key role in TCR assembly and activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested with normal human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample to antibody cocktails prior to combining with cells.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. CD3 gamma, delta, epsilon, and zeta subunits are structurally related immunoglobulin superfamily members encoded on chromosome 11. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes but not on B or NK cells. Defects in CD3 genes can lead to immunodeficiency.
