Product Specific Information

The OKT3 monoclonal antibody binds to an epitope on the epsilon-subunit of the human CD3 complex. It exhibits potent immunosuppressive properties and is effective in treating renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by developing thymocytes and all mature T cells. Crosslinking of TCR triggers intracellular signaling leading to activation and proliferation. Clones OKT3 and SK7 recognize different epitopes.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.6 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Note:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits—members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with cytoplasmic signaling molecules through tyrosine-based motifs. CD3 plays roles in TCR-mediated activation, proliferation, and survival.

• Expression: 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum
• Not expressed on: B or NK cells
• Clinical relevance: Decreased T lymphocytes may occur in autoimmune diseases.
• Genetic information: CD3 epsilon, gamma, and delta genes are located on chromosome 11; defects can cause CD3 immunodeficiency.
