Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex.
It has potent immunosuppressive properties and has been proven effective in the treatment of organ transplant rejection (renal, heart, liver).

CD3 subunits (gamma, delta, epsilon) are required for proper assembly, trafficking, and surface expression of the TCR complex.
CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR initiates intracellular activation and proliferation pathways.
Antibody clones OKT3 and SK7 recognize different epitopes.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.6 µg)/test.
A test is defined as staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample (regardless of number of NovaFluors)
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression.
CD3 gamma, delta, epsilon, and zeta subunits are structurally related members of the immunoglobulin superfamily encoded on chromosome 11.
These subunits associate with signal transduction molecules via tyrosine-based motifs.
CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum.
Not expressed on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
