Target Information
ERO1L (endoplasmic oxidoreductin-1-like protein) is localized in the ER membrane as a peripheral membrane protein on the lumenal side. Its association with ERP44 is essential for retention in the ER and for coupling oxygen reduction to de novo disulfide formation.
ERO1L is broadly distributed at low levels in several tissues, with highest levels in the upper digestive tract, and is stimulated by hypoxia via HIF signaling.
It generally exists as a monomer but can function as a homodimer and interacts with PDILT. After translation, ERO1L becomes N-glycosylated, and its disulfide bonds constitute the redox-active center: Cys-94/Cys-99 accepts an electron from P4HB, funneling it to the active site disulfide Cys-394/Cys-397.
ERO1L oxidizes P4HB/PDI isomerase through direct disulfide exchange and associates with ERP44. It plays a key role in immunoglobulin folding, oxidative stress response, release of unfolded cholera toxin upon Vibrio cholerae infection, and ER stress-induced CHOP-dependent apoptosis via IP3R1 activation.
