Target Information
This gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which are structurally related isoforms functioning as inhibitors of CDK4 kinase. Another transcript contains an alternate first exon located 20 Kb upstream and encodes a structurally unrelated ARF protein that stabilizes tumor suppressor p53 by interacting with and sequestering MDM2, the E3 ubiquitin-protein ligase responsible for p53 degradation. Despite structural differences, CDK inhibitor isoforms and the ARF product share a common role in G1 cell cycle control. This gene is frequently mutated or deleted in various tumors and serves as an important tumor suppressor gene.
