Target Information
ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration. The Rho/ROCK pathway is involved in the progression of various human cancers. ROCK1 has been identified as a Caspase-3 substrate.
ROCK, an effector of the small GTPase Rho, contributes to myosin light chain (MLC) activation through direct phosphorylation of MLC and inhibition of MLC phosphatase.
During apoptosis, ROCK1 is cleaved by caspase-3 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity. This cleavage induces MLC phosphorylation and membrane blebbing, key events in apoptosis execution.
