ATG5 (Autophagy Related 5)는 오토파지 과정의 핵심 요소로, 세포사멸(apoptosis)에도 중요한 역할을 합니다.
이 단백질은 미토콘드리아 품질 관리, 항바이러스 면역 반응의 음성 조절, 림프구 발달 및 증식, MHC II 항원 제시, 지방세포 분화 등 다양한 세포 과정에 관여합니다.
ATG12와 결합한 ATG5-ATG12 복합체는 오토파고좀 형성에 참여하며, FADD 단백질과 상호작용하여 오토파지성 세포사멸을 유도합니다.
또한, RIG-I 및 VISA(IPS-1)와 같은 선천 면역 단백질과 결합하여 제1형 인터페론 생성을 억제하고 바이러스 복제를 허용합니다.
ATG5 기능 이상은 소뇌실조증(spinocerebellar ataxia)과 관련이 있습니다.
