Target Information
The capsid protein self-assembles to form an icosahedral capsid with T=1 symmetry, approximately 22 nm in diameter, composed of 60 copies of three variants (VP1, VP2, VP3) differing in their N-terminus.
The capsid encapsulates the genomic ssDNA and binds to host cell heparan sulfate, using host ITGA5-ITGB1 as a coreceptor for virion attachment. This interaction induces internalization predominantly via clathrin-dependent endocytosis.
Binding to the host receptor triggers capsid rearrangements, exposing the VP1 N-terminus with its phospholipase A2-like region and potential nuclear localization signals. The VP1 N-terminus may act as a lipolytic enzyme to breach the endosomal membrane and facilitate viral transport to the nucleus.
