Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is Alzheimer’s brain tissue.

Target Information

Amyloid beta peptide (Abeta/Beta-amyloid) is the major constituent of amyloid plaques in the brains of individuals afflicted with Alzheimer’s disease.
Abeta peptide (40–43 amino acids) is generated from the beta-amyloid precursor protein (beta APP) through a two-step cleavage process involving beta-secretase (BACE) and gamma-secretase.
Gamma-secretase, a multi-subunit complex containing presenilin-1 and -2, cleaves the carboxyl-terminal domain of beta APP to generate amyloid beta peptide.
Abeta is an extracellular filamentous protein component of amyloid cores and neuritic plaques and is also found in neurofibrillary tangles.
Beta amyloid deposits are observed in Alzheimer’s disease, Lewy body dementia, Down’s syndrome, amyloidosis (Dutch type), cerebroarterial amyloidosis, and the Guam Parkinson-Dementia complex.
