Target Information
Hyaluronic acid (HA) is a high molecular weight unbranched polysaccharide present in the extracellular matrix of many organisms, from bacteria to mammals. It consists of alternating glucuronic acid and N-acetylglucosamine residues linked by β-1,3 and β-1,4 glycosidic bonds. HA is synthesized by membrane-bound synthases at the inner surface of the plasma membrane and extruded into the extracellular space.
HA functions in space filling, joint lubrication, and as a matrix for cell migration. It is actively produced during wound healing and tissue repair, providing a scaffold for blood vessel and fibroblast ingrowth. Altered HA levels are associated with inflammatory and degenerative arthropathies such as rheumatoid arthritis. The interaction of HA with CD44 on leukocytes is crucial for tissue-specific homing, and overexpression of HA receptors correlates with tumor metastasis.
HAS1 belongs to a vertebrate gene family encoding hyaluronan synthases and shows significant homology to the hasA gene product of Streptococcus pyogenes.
