Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a (LAMP-1), a type I lysosomal membrane protein that is heavily glycosylated and expressed in late endosomes and lysosomes. CD107a transiently appears on the surface of degranulating cytolytic T cells and is associated with processes such as cancer metastasis and lysosomal storage disorders.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis
  • Surface staining in flow cytometric degranulation assays

Applications Tested:

  • Tested by intracellular staining and flow cytometric analysis of mouse thioglycolate-elicited peritoneal exudate cells
  • Recommended ≤ 1 µg/test (in 100 µL final volume)
  • Cell number range: 10⁵–10⁸ cells/test
  • Optimize antibody titration for best performance

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Target Information

LAMP1 (CD107a, lysosome-associated membrane protein-1) is a major lysosomal membrane component. About 1–2% is found on the plasma membrane. It is a heavily glycosylated type I integral membrane protein involved in lysosome biogenesis and fusion with phagosomes. LAMP1 translocates from the trans-Golgi network to endosomes and lysosomes. Upon activation, LAMP1 moves to the plasma membrane via a tyrosine-based motif (YXXI). Mutations disrupting this motif affect lysosomal localization.

LAMP1 and LAMP2 promote adhesion of human PBMCs to vascular endothelium and may play roles in inflammation. Increased LAMP1 expression is observed in neurons and glial cells around senile plaques in Alzheimer’s disease and in thymic epithelial cells and macrophages during acute thymic involution. LAMP1 serves as a marker of mast cell activation.
