Description:
This MIH64 monoclonal antibody recognizes mouse VISTA (also known as B7-H5 or PD-H1). The antibody is validated for flow cytometry on both native and paraformaldehyde-fixed cells.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes at ≤ 0.5 µg/test (defined as the amount of antibody staining a cell sample in 100 µL final volume). Recommended to titrate for optimal performance. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor™ 710 emits at 710 nm and is excited with the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be verified empirically.

Target Information

VISTA (V-domain Ig containing suppressor of T-cell activation)
Also known as B7-H5 or PD-1H, encoded by the VSIR gene.

• Shares sequence homology with PD-L1 but differs in expression pattern.
• Expressed predominantly on hematopoietic cells (highest on myeloid cells, lower on T cells, absent on B cells).
• VISTA on antigen-presenting cells suppresses proliferation and cytokine production of CD4/CD8 T cells.
• Important target for cancer immunotherapy.
• Known ligands: CD28H, VSIG-3, VSIG-8.
