Target Information

ACY1 encodes a cytosolic, homodimeric, zinc-binding enzyme that catalyzes the hydrolysis of acylated L-amino acids to L-amino acids and an acyl group. It is involved in the catabolism and salvage of acylated amino acids. The gene is located on chromosome 3p21.1, a region often reduced to homozygosity in small-cell lung cancer (SCLC), where its expression is frequently diminished or absent. The human aminoacylase-1 sequence shows high homology with the porcine enzyme and represents the first member of a new zinc-binding enzyme family. Mutations in ACY1 cause aminoacylase-1 deficiency, a metabolic disorder characterized by CNS defects and elevated urinary N-acetylated amino acids. Multiple transcript variants exist due to alternative splicing, and read-through transcription occurs with the upstream ABHD14A gene. A related pseudogene is found on chromosome 18.
