Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (Fc gamma RIII), which exists as two isoforms: FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, and NK cells.
• FcγRIIIB: Expressed on neutrophils as a GPI-anchored form.

In non-human primates, CD16 expression patterns differ slightly; for example, neutrophils in macaques and baboons lack CD16, while sooty mangabeys express it.

Binding of IgG to CD16 triggers signal transduction leading to ADCC, phagocytosis, cytokine release, and antigen presentation.

• Inhibition of cytotoxic ability
• Activation of signaling pathways
• NK cell depletion in vivo

Cross-reactivity: Works on multiple primate species including capuchin monkey, chimpanzee, marmoset, baboon, macaque, rhesus, and squirrel monkey.
Epitope: Recognizes same epitope as CB16; distinct from B73.1.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C with minimal brightness loss.
