Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) that detects viral double-stranded RNA (dsRNA), initiating innate immune responses. Together with MDA5, RIG-I activates antiviral signaling pathways by recognizing specific RNA structures. RIG-I primarily detects short dsRNAs lacking a 5'-triphosphate end, while MDA5 recognizes long dsRNAs. Upon activation, both signal through IPS-1 to activate NF-κB and IRF-3, leading to cytokine production, inflammation, and apoptosis. RIG-I plays a critical role in immune responses to viruses such as influenza A and B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. It is also involved in cytosolic DNA detection via RNA polymerase III. MicroRNA-146a is known to negatively regulate RIG-I–dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
