Target Information
IGF1R (Insulin-like Growth Factor 1 Receptor) is a receptor tyrosine kinase activated by IGF-1 and IGF-2.
It plays a role in various cancers. The protein forms a tetramer of two alpha and two beta subunits.
Alpha-subunits bind IGF-1 with higher affinity than IGF-2, inducing conformational change and autophosphorylation at residues 1134, 1135, and 1136.
Activated IGF-1R phosphorylates substrate proteins, triggering RAS/RAF/MAPK pathways (ERK, p38, JNK) that promote gene transcription and cell proliferation.
