Target Information
The INK4a locus on chromosome 9p21 is frequently affected in human tumors. It encodes two structurally distinct tumor suppressor proteins, p16INK4a and ARF (p14ARF in human, P19ARF in mouse). These proteins interact with retinoblastoma protein and p53 tumor suppressors, respectively, playing key roles in cell senescence, cell cycle control, and tumor suppression. p14ARF can induce both G1 and G2 phase arrest in a p53-dependent manner. Functional differences between mouse and human ARF proteins may contribute to species-specific tumor susceptibility.
