Product Specific Information

The monoclonal antibody 3DS304M recognizes mouse CD304 (Neuropilin-1), a type 1 transmembrane glycoprotein expressed on multiple cell types including regulatory T cells (Treg), NKT cells, dendritic cells, certain stem cells, neurons, endothelial cells, and some neoplastic cells.

Neuropilin-1 functions as a co-receptor for type 3 semaphorins in growing neurons and participates in angiogenesis as part of the VEGF receptor complex in endothelial cells. In mice, Neuropilin-1 is expressed on thymus-derived natural Treg but not peripherally induced Treg. Transient expression has also been observed on recently activated non-regulatory T cells.

This antibody recognizes formaldehyde-fixed and methanol-treated epitopes.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Tested on mouse splenocytes at ≤0.06 µg/test (100 µL assay volume). Optimize titration for best performance.
  • Fluorochrome: PerCP-eFluor® 710 (Emission 710 nm, Excitation 488 nm, Blue laser)
    • Recommended filter: 710/50 bandpass (695/40 acceptable)

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Neuropilin 1 (NRP1) is a 923-amino-acid membrane-bound co-receptor belonging to the neuropilin family, containing two CUB domains, two F5/8 type C domains, and one MAM domain.

It forms heterodimers with Neuropilin 2 (NRP2) and binds plexin (PLXNB1) to form semaphorin receptors. Predominantly expressed in the nervous system, Neuropilin 1 is involved in cardiovascular development, angiogenesis, axon guidance, neuronal circuit formation, cell survival, and organogenesis.

It acts as a receptor for both semaphorins and VEGF and regulates VEGF-induced angiogenesis. Neuropilin 1 has been implicated in advanced prostate carcinoma and breast cancer progression.
