The Human TNF-RI Simplex ProcartaPlex Kit measures TNF-RI protein and is designed to be combinable with other Simplex kits, allowing you to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously on a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and yield analyzed results in approximately four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single small-volume sample from plasma, serum, cell culture supernatants, or other fluids.

Each Luminex bead in the ProcartaPlex assay is internally dyed with specific ratios of red and infrared fluorophores, creating unique spectral signatures identifiable by Luminex systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX). Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each spectrally unique bead is labeled with antibodies specific for a single target, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed onto a microplate well, allowing free-floating assay reagents in solution. Detection instruments such as Luminex 200 use two lasers—one to identify bead signatures and another to quantify RPE fluorescence, proportional to the protein concentration. This enables profiling of more targets using less sample and time compared to conventional ELISA.

ProcartaPlex multiplex panels are available across six species (human, mouse, rat, nonhuman primate, porcine, and canine).
