The Human ICAM-1 Simplex ProcartaPlex Kit measures ICAM-1 protein and is designed to be combinable with other Simplex kits to create custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (i.e., not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of a protein to quantitate multiple targets simultaneously on a Luminex instrument. These multiplex assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and deliver analyzed results within four hours.

ProcartaPlex assays use Luminex xMAP (multianalyte profiling) technology for the simultaneous detection and quantitation of up to 80 protein targets in a single sample. The Luminex beads are internally dyed with specific ratios of red and infrared fluorophores to create unique spectral signatures identifiable by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, matched antibody pairs identify the target protein. Each spectrally unique bead is labeled with antibodies specific for one protein, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The bead-conjugated capture antibodies enable simultaneous analysis of multiple targets in a single well.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed to the microplate well, allowing reagents to remain in solution. Luminex instruments use dual lasers—one to identify the bead and another to quantify RPE fluorescence, which correlates with protein concentration. This enables profiling of more proteins using less sample within the same assay time.
