Product Specific Information

The HM40-3 monoclonal antibody reacts with mouse and rat CD40, a 45–50 kDa type I transmembrane glycoprotein belonging to the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and certain peripheral T cells. It regulates B cell development and maturation, induces Ig isotype switching, and protects B cells from apoptosis when combined with IL-4 signaling. Interaction with CD154 (gp39) on T cells is critical for T-B cell communication and immune regulation.

Target Information

CD40 is a single-chain glycoprotein and a member of the TNF/NGF superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and dendritic cells. CD40 is essential for immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. It interacts with TNFR2 and CD40 ligand, playing a role in microglial activation and Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
