Target Information
Ago proteins are broadly expressed in somatic cells, associate with miRNAs, and are key actors in various RNA silencing pathways. Among them, only Ago2 displays endonucleolytic or “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when base-pairing is perfect. With partial complementarity, Ago2 inhibits translation through repression rather than cleavage.
Ago2 plays a non-redundant role in small RNA-guided gene silencing, translation repression, and heterochromatinization. As a core component of the RISC complex, AGO2 catalyzes degradation of target mRNAs guided by siRNAs or miRNAs. It also functions as an RNA slicer independently of Dicer and regulates miRNA maturation.
Overexpression of Ago2 has been linked to tumor cell growth and cancer progression. Mouse gene disruption studies show Ago2 is essential for embryonic development and regulates B-lymphoid and erythroid differentiation.
