Target Information
Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in the detection of viral double-stranded RNA (dsRNA). Along with MDA5, RIG-I activates the innate immune response by recognizing viral RNA.
RIG-I responds to short dsRNAs without a 5'-triphosphate end, while MDA5 responds to long dsRNAs. Both signal through IPS-1, activating NF-kappaB and IRF-3, leading to apoptosis, cytokine signaling, and inflammation.
RIG-I plays a critical role in the immune response to influenza A/B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus.
MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
Recent studies suggest RIG-I also detects cytosolic DNA via RNA polymerase III activity.
