GAD-65와 GAD-67은 group II decarboxylase 단백질군에 속하며, L-glutamic acid로부터 GABA(gamma-aminobutyric acid)를 생성하는 과정의 속도 제한 단계를 촉매합니다.
GAD-65는 시냅스 소포막에 주로 존재하며, GAD-67은 세포 전반에 분포합니다. GAD-67은 기본적인 GABA 합성에 관여하며, 신경전달 시 GABA 수요가 증가할 경우 GAD-65가 일시적으로 활성화되어 GABA 생산을 돕습니다.
GAD-65의 손실은 GABA 신경전달에 손상을 주며, GAD-67의 손실은 치명적입니다. GAD-67에는 대체 스플라이싱으로 인한 두 가지 isoform(GAD-67 및 GAD-25)이 존재합니다.
