Target Information
Voltage-gated potassium (Kv) channels are essential components of potassium currents in the heart and central nervous system.
In the heart, Kv4 channels participate in the repolarization phase of the action potential, while in the brain they prevent reverse-propagation of action potentials.
Kv4 channels are associated with calcium-binding proteins known as KChIPs (Kv channel interacting proteins), which modulate channel function.
There are three known KChIPs:
- KChIP1: Expressed in the brain
- KChIP2: Expressed in brain, heart, and lung (three isoforms: a, b, c)
- KChIP3: Also known as calsenilin or DREAM (DRE-antagonist modulator), expressed in brain and testis
KChIP3 not only modulates Kv4 currents but also binds to regulatory DRE elements and represses transcription of the early-response gene c-fos, functioning as a calcium-binding transcriptional regulator.
It is also associated with presenilins and implicated in beta-amyloid formation.
