Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53. It plays a key role in the cellular response to genotoxic stress and acts as a major transcriptional target of p53. Nuclear p21 binds and inhibits Cdk1 and Cdk2, blocking the transition from G1 to S phase or from G2 to mitosis after DNA damage, allowing DNA repair. In the cytoplasm, p21 has anti-apoptotic effects by inhibiting caspase 3 and apoptotic kinases ASK1 and JNK. p21 exhibits dual roles in carcinogenesis, functioning as a tumor suppressor and, paradoxically, as an oncogene under certain conditions.
