• For IHC(P), use heat-induced antigen retrieval in pH 6.0 citrate buffer.

Target Information

Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
This protein is a key component of the extracellular matrix in cartilage, providing resistance to compression.
Human Aggrecan (~250 kDa) consists of 2316 amino acids and multiple isoforms due to alternative splicing.
It exhibits a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to a protein core.
Aggrecan synthesis and degradation are studied for their roles in cartilage deterioration during joint injury, disease, and aging.
It contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.
Aggrecan fragments are released into synovial fluid during all stages of osteoarthritis due to cleavage by aggrecanase and MMP activity.
Antibodies BC-3 and BC-4 detect neo epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
