Target Information
STAT3 (Signal Transducer and Activator of Transcription 3)는 STAT 단백질 패밀리에 속하며, receptor-associated kinase에 의해 인산화되어 핵으로 이동 후 전사인자로 작용합니다. STAT3는 cytokines 및 growth factors에 반응하여 활성화되며, CREB-binding protein과 같은 coactivator가 필요합니다.
Interferon-alpha, Interferon-gamma, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, tyrosine 705의 JAK1 및 JAK2 인산화가 dimer 형성, 핵 이동, DNA 결합에 필수적입니다.
STAT3 유전자는 인간 염색체 17q에 위치하며, IFN-alpha에 의해 활성화되지만 IFN-beta에는 반응하지 않습니다. c-Jun 및 CREB와 결합하여 다양한 유전자 발현을 매개하며, 세포 성장과 apoptosis 등 여러 세포 과정에 중요한 역할을 합니다. Rac1은 STAT3의 활성을 조절하고, PIAS3는 STAT3의 특이적 억제 단백질입니다.
STAT3는 세 가지 alternative splicing isoform을 가지며, knockout mouse에서는 배아 초기 단계에서 치명적입니다.
