Target Information
CD40 항원은 tumor necrosis factor/nerve growth factor superfamily의 단일 사슬 당단백질로, Hodgkin's disease 관련 항원 CD30과 높은 상동성을 보입니다.
CD40은 plasma cell을 제외한 모든 B 세포, 일부 상피세포, 암종 및 림프성 수지상세포에서 발현됩니다.
T 세포 의존성 면역글로불린 클래스 전환, 메모리 B 세포 발달, germinal center 형성 등 다양한 면역 및 염증 반응에 필수적인 역할을 합니다.
또한 CD40과 TNFR2의 상호작용은 신호전달 매개체로 작용하며, CD40–CD40L 결합은 알츠하이머병 병리학의 초기 단계로 알려진 amyloid-beta 유도 미세아교세포 활성화에 필요합니다.
두 가지 대체 스플라이싱 전사체가 보고되어 있으며, CD40 기능 이상은 Type 3 Hyper-IgM immunodeficiency 및 CD40 ligand deficiency와 관련됩니다.
