Target Information
MMP (matrix metalloproteinase)는 결합조직 성분을 분해할 수 있는 단백질분해 효소군으로, 공통적인 활성화 메커니즘과 금속 결합 부위의 보존된 아미노산 서열을 가지고 있습니다. 이들은 TIMPs(tissue inhibitors of metalloproteinases)에 의해 억제됩니다. MMP와 TIMP는 혈관신생 조절에 중요한 역할을 합니다.
MMP2는 631 아미노산으로 구성된 전구 효소(proenzyme) 형태로 합성되며, N-말단의 80 아미노산 절단을 통해 활성화됩니다. Propeptide, catalytic, hemopexin 도메인으로 구성되어 있습니다.
MMP 계열은 최소 18종 이상의 분비형 및 막결합형 zinc endopeptidase로 구성되어 있으며, 세포외기질의 모든 주요 성분(콜라겐, 피브로넥틴, 라미닌 등)을 분해할 수 있습니다.
MMP2는 조직 재형성에 관여하며, MMP-2 유전자의 돌연변이는 Winchester syndrome 및 Nodulosis-Arthropathy-Osteolysis(NAO) syndrome과 관련이 있습니다. 두 가지 전사체 변이가 알려져 있습니다.
