Target Information
ROCK1 is a serine/threonine kinase that regulates cell differentiation and migration.
The Rho/ROCK pathway is involved in the progression of various human cancers.
ROCK1 has been identified as a Caspase-3 substrate. It contributes to myosin light chain (MLC) activation via direct phosphorylation of MLC and phosphorylation of MLC phosphatase, leading to its inhibition.
This results in increased cellular contractility necessary for apoptotic membrane blebbing.
ROCK1 consists of an amino-terminal kinase domain and a cysteine/histidine-rich C-terminal domain within a pleckstrin-homology region, connected by a Rho-binding domain.
During apoptosis, ROCK1 is cleaved by caspase-3 at a conserved DETD1113/G sequence, removing its inhibitory domain and resulting in constitutive kinase activity.
This cleavage induces phosphorylation of MLC and membrane blebbing, a key event in the execution phase of apoptosis.
