Target Information

Activator of S phase kinase (ASK, DBF4) is a regulatory subunit of the human Cdc7 kinase complex that plays a pivotal role in the G1/S cell cycle transition in mammalian cells. The increase in Cdc7 kinase activity at the G1/S boundary is partly due to elevated ASK expression in late G1. ASK interacts with replication origins in vivo, suggesting that Cdc7 may trigger S phase by activating replication initiation complexes. Human ASK protein levels are regulated during the cell cycle, matching the pattern of human Cdc7 kinase activity. The Cdc7-ASK kinase complex is directly involved in regulating DNA replication initiation by targeting MCM2 protein. Nuclear localization and chromatin binding of huCdc7 and GFP-ASK occur through nuclear localization signals at telophase, possibly requiring additional signals for chromatin binding. ASK may also functionally interact with menin, a tumor suppressor mutated in MEN1, in the regulation of cell proliferation.
