- Positive Control: Ramos cells serum-starved for 16 hours, then treated with 12 µg/mL human IgM (diluted in serum-free medium) for 2 minutes.

Target Information
Spleen tyrosine kinase (SYK) is a cytoplasmic tyrosine kinase, similar to ZAP70, involved in immune receptor signaling. It translocates to the plasma membrane upon activation of the B cell antigen receptor (BCR) or high-affinity IgE receptor (FcεRI), phosphorylating downstream adaptor proteins and initiating signaling pathways such as calcium mobilization, cytoskeleton remodeling, and gene transcription. SYK interacts with receptor ITAM motifs phosphorylated by Src-family kinases, which also activate SYK by phosphorylation of its activation loop. SYK is considered a potential drug target for allergic and autoimmune diseases and may also be involved in cancer.
