Product Specific Information

The OX35 monoclonal antibody reacts with rat CD4, a co-receptor involved in T cell receptor signaling via MHC class II on antigen-presenting cells. CD4 is expressed by CD4+CD8+ double-positive and CD4+ single-positive thymocytes, as well as CD4+ helper T cells.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Flow cytometric analysis of rat splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration recommended.
• Fluorochrome: Super Bright 780 (excited at 405 nm, emits at 780 nm; use 780/60 bandpass filter).

Experimental Notes:

• Compensation values may differ between beads and cells; recommended to set compensation with cells.
• When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (SB-4401) to minimize non-specific interactions.
• Light-sensitive dye: protect from light.
• Fixation: Samples can be stored up to 3 days at 4°C in IC Fixation Buffer or 1-step Fix/Lyse Solution with minimal impact on brightness.
• Laser: Violet (405 nm).

Target Information

CD4 antigen recognizes MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed in T-helper lymphocytes, and also found in monocytes, macrophages, and dendritic cells. CD4 expression occurs in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut. CD4 participates in early T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
