• Immunohistochemistry (PFA-fixed): Incubate antibody for 30 min at room temperature.
• For staining of formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on the surface of dendritic cells and macrophages. It plays a critical role in the innate immune system, recognizing a wide range of pathogens from parasites to viruses.
The protein comprises three domains:

1. N-terminal transmembrane domain
2. Tandem-repeat neck domain
3. C-type lectin carbohydrate recognition domain

The extracellular region (neck + lectin domains) functions as both a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands on microbial and host cell surfaces. The neck region enables homo-oligomerization, allowing high-avidity binding to multivalent ligands.
Variations in the number of 23 amino acid repeats in the neck domain can significantly affect ligand-binding capability.
DC-SIGN is closely related to L-SIGN (GeneID 10332), differing in ligand-binding properties and tissue distribution. Alternative splicing produces multiple isoforms.
