Target Information

This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on dendritic cells and macrophages. It functions in the innate immune system by recognizing diverse pathogens including parasites and viruses.
The protein has three domains:

  • N-terminal transmembrane domain
  • Tandem-repeat neck domain
  • C-type lectin carbohydrate recognition domain

The extracellular region (neck + lectin domains) acts as both a pathogen recognition receptor and a cell adhesion receptor by binding carbohydrate ligands. The neck region enables homo-oligomerization, enhancing multivalent ligand binding.
Rare variations in the number of 23 amino acid repeats in the neck domain can significantly impact ligand binding ability. This gene is closely related to L-SIGN (GeneID 10332), differing in ligand-binding properties and tissue distribution.
Alternative splicing results in multiple variants.
