Target Information

RIPK2 is a serine/threonine kinase involved in cell activation, proliferation, differentiation, and apoptosis. Upon stimulation by bacterial peptidoglycans, NOD1 and NOD2 are activated, oligomerize, and recruit RIPK2 through CARD-CARD domains. RIPK2 contributes to tyrosine phosphorylation of ARHGEF2 via Src kinase, leading to NF-κB activation. Once recruited, RIPK2 autophosphorylates and undergoes Lys-63-linked polyubiquitination by E3 ligases XIAP, BIRC2, and BIRC3. This modification facilitates MAP3K7/TAK1 recruitment to IKBKG/NEMO, inducing NF-κB activation and transcription of genes related to immune response and apoptosis. RIPK2 also promotes BCL10 phosphorylation during T-cell receptor engagement.
