Product Specific Information

The eBio133-1C6 monoclonal antibody reacts with human CD100, a member of the class IV semaphorin family also called sema4D.
CD100 promotes angiogenesis, invasive growth, proliferation, and anti-apoptosis of cancer cells in vitro. It acts as a costimulatory molecule in adaptive immune responses by switching off negative signalling from CD72. Another ligand is Plexin-B1.
Higher expression levels of CD100 are correlated with poor survival in soft tissue sarcoma patients.
Expression is found in both hematopoietic and non-hematopoietic cells, including resting T cells, NK cells, and B cells. CD100 can also exist as a soluble form.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry on human peripheral blood cells.
Use at 5 µL (0.25 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Semaphorins are neuronal chemorepellants that guide neurons during development.
CD100 (Semaphorin 4D), a 150 kDa homodimer, induces B cell aggregation and improves viability in vitro.
It modifies CD40-CD40L signaling by enhancing B cell aggregation and survival and downregulating CD23 expression.
CD72 is a lymphocyte receptor for CD100, regulating B cell signaling.
CD100 is expressed on resting and PHA-stimulated T cells, weakly on NK cells, EBV-transformed B cells, monocytes, and tumor/peripheral blood T cell lines, and at higher density on activated T and B cells.
