Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and plays a vital role in brain development. Mutations, rearrangements, and amplifications in the ALK gene have been identified in tumors such as anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement is the most common genetic alteration, creating a fusion gene between ALK and nucleophosmin (NPM). The NPM-ALK fusion protein is oncogenic. ALK encodes a receptor tyrosine kinase with transmembrane and extracellular domains, absent in the NPM-ALK product, and shows high sequence similarity to LTK.
ALK is essential for neuronal development and function in the nervous system.
