Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising about 10% of all surface proteins in lymphocytes. It is absent in non-hematopoietic tissues and cell lines.
CD45 plays a critical role in lymphocyte development and antigen signaling by regulating Src-family kinases. Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
CD45RA, one isoform, shows restricted expression among lymphoid cell subtypes. CD45 is essential for T- and B-cell receptor signaling and modulates cytokine receptor pathways through suppression of JAK kinases. It also influences cell survival, apoptosis, and integrin-mediated signaling.
