Target Information
RIPK3 is a member of the receptor-interacting protein (RIP) family of serine/threonine protein kinases, which contains a C-terminal domain unique from other RIP family members. RIPK3 is predominantly localized to the cytoplasm and is colocalized in the mitochondrion, capable of nucleocytoplasmic shuttling dependent on nuclear localization and export signals. It is part of the TNF receptor-I signaling complex, can induce apoptosis, and weakly activates NF-kappaB. Caspase-8 prevents RIPK3-dependent necrosis by forming a proteolytically active complex with CFLAR, which is required for its protective function.
