Product Specific Information

The monoclonal antibody 162 recognizes CRACC (CD2-like receptor for activating cytotoxic cells), also known as CD319 and CS1, a member of the SLAM receptor family.
CRACC is a 66 kDa glycosylated transmembrane protein containing a cytoplasmic ITSM domain (Immunoreceptor Tyrosine-based Switch Motif). This domain interacts with SH2-containing proteins such as SAP, but in CRACC, signaling is SAP-independent and mediated by EAT-2.
A splice variant lacking the ITSM domain has been reported.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated on normal human peripheral blood cells.
Recommended usage: 5 µL (0.5 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Use 710/50 bandpass filter (695/40 acceptable alternative). Ensure instrument compatibility.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333).
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

SLAMF7 contains one Ig-like C2-type domain. Isoform 1 mediates NK cell activation via SAP-independent ERK pathway and may aid lymphocyte adhesion.
Isoform 3 does not mediate activation. SLAMF7 is expressed in spleen, lymph node, peripheral blood leukocytes, bone marrow, and various tissues.
Detected in NK cells and activated B-cells but not in promyelocytic, B-, or T-cell lines. Isoform 3 is expressed at much lower levels than isoform 1.
Three named isoforms are known.
