Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen, also known as BCM1, Blast-1 (human), and OX-45 (rat). CD48 is a 45 kDa GPI-linked glycoprotein of the SLAM and Ig superfamily, expressed on most hematopoietic cells.

CD48 expression patterns among hematopoietic progenitors can indicate stem cell primitiveness:

• HSC: CD150(+)CD244(-)CD48(-)
• MPP: CD244(+)CD150(-)CD48(-)
• Restricted progenitors: CD48(+)CD244(+)CD150(-)

CD48 plays a critical role in adhesion and T cell activation. In mice, its counter-receptors are CD2 and CD244. HM48-1 can modulate CD48 functions, blocking CD48/CD2 and CD48/CD244 interactions, inhibiting spleen cell proliferation, providing costimulation for T cells, and prolonging cardiac allograft survival.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.125 µg/test, 100 µL final volume). Cell numbers: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Buffer Usage:
Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining:

• Add 5 µL per sample to antibody cocktail before mixing with cells.
• For single-color controls: 5 µL per 100 µL cell sample.

NovaFluor conjugates utilize Phiton™ technology, based on engineered nucleic acid dye structures for optimized fluorescence with minimal spillover and spread.

Target Information

CD48 (Blast-1) is a GPI-anchored protein of the CD2 subset of the Ig superfamily. It mediates homotypic and heterotypic adhesion through interactions with CD2 and 2B4. CD48 is broadly expressed on hematopoietic cells and regulates NK and NK-T cell proliferation.

• Strongly expressed: lymphocytes, monocytes
• Weakly expressed: granulocytes
• Absent: platelets, fibroblasts, epithelium, endothelium

CD48 is a marker for detecting GPI anchoring defects (e.g., paroxysmal nocturnal hemoglobulinuria, PNH) and may serve as a pharmacological target in allergic airway diseases such as rhinitis and asthma.
