Product Specific Information

The HM48-1 monoclonal antibody reacts with mouse CD48 antigen (also known as BCM1, Blast-1 in human, and OX-45 in rat).
CD48 is a 45 kDa GPI-linked glycoprotein of the SLAM family, expressed on most hematopoietic cells. It serves as a useful marker for identifying hematopoietic progenitor populations, including HSCs (CD150⁺CD244⁻CD48⁻), MPPs (CD244⁺CD150⁻CD48⁻), and restricted progenitors (CD48⁺CD244⁺CD150⁻).

CD48 plays a critical role in adhesion and T cell activation. In mice, its primary counter-receptors are CD2 and CD244. HM48-1 modulates CD48 functions in vitro and in vivo, including:

  • Blocking CD48/CD2 and CD48/CD244 interactions
  • Inhibiting mitogen-activated spleen cell proliferation
  • Providing costimulation for TCR-activated T cells
  • Prolonging cardiac allograft survival in vivo

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.2 µg/test).
A test corresponds to staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Important Note:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample (regardless of number of NovaFluors).
For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology using novel nucleic acid dye structures for engineered fluorescent signatures optimized for minimal spillover and spread.

Excitation: 636 nm
Emission: 756 nm
Recommended Laser: 633–640 nm (Red)


Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily (CD2, CD58, CD84, CD150, CD229, CD244, etc.).
It mediates homotypic and heterotypic adhesion through interactions with other Ig family members.
CD48 is GPI-anchored and broadly expressed on hematopoietic cells, serving as:

  • High-affinity ligand for 2B4
  • Low-affinity ligand for CD2

The 2B4-CD48 interaction regulates NK and NK-T cell proliferation. CD48 signaling activates eosinophils and is upregulated in infectious diseases.
Expression pattern:

  • Strong: lymphocytes, monocytes
  • Weak: granulocytes
  • Absent: platelets, fibroblasts, epithelium, endothelium

CD48 is a marker for detecting GPI anchoring defects (e.g., paroxysmal nocturnal hemoglobulinuria, PNH) and may be involved in allergic airway disease pathology, offering potential pharmacological targets for conditions such as allergic rhinitis and asthma.
