Product Specific Information

The HM48-1 monoclonal antibody reacts with the mouse CD48 antigen (also known as BCM1, Blast-1 in human, and OX-45 in rat). CD48, a 45 kDa GPI-linked glycoprotein in the SLAM and Ig superfamily, is expressed on most hematopoietic cells. It plays a critical role in adhesion and T-cell activation.
In mouse, CD48 interacts with CD2 and CD244. HM48-1 modulates CD48 functions including blocking CD48/CD2 and CD48/CD244 interactions, inhibiting mitogen-activated spleen cell proliferation, providing costimulation for T cells, and prolonging cardiac allograft survival.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; recommended ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily, including CD2, CD58, CD84, CD150, CD229, and CD244. These molecules mediate homotypic or heterotypic adhesion.
CD48 is GPI-anchored and broadly expressed on hematopoietic cells, serving as a high-affinity ligand for 2B4 and low-affinity ligand for CD2.
2B4–CD48 interaction among NK and NK-T cells regulates proliferation. CD48 signaling activates eosinophils and is upregulated in several infectious diseases.
CD48 is strongly expressed on lymphocytes and monocytes, weakly on granulocytes, and absent on platelets, fibroblasts, epithelium, and endothelium.
It is a marker for detecting GPI-anchor defects in paroxysmal nocturnal hemoglobinuria (PNH) and may serve as a pharmacological target in allergic airway diseases such as rhinitis and asthma.
