Target Information
Zero, also known as myelin protein zero (MPZ), is a Type I integral membrane glycoprotein that mediates adhesion of spiraling wraps of the myelin sheath to ensure stable synaptic transmission.
MPZ constitutes approximately 50% of total protein in the myelin sheath, contributing to structural integrity of peripheral myelin.
It guides compact myelin wrapping through glycine zipper packing interface-dependent dimer and tetramer formation.
Mutations (e.g., G134R) can disrupt multimer formation and cause demyelinating neuropathies such as Charcot-Marie-Tooth disease.
The cytoplasmic domain undergoes serine and tyrosine phosphorylation, which is prevalent during peak nerve myelination.
MPZ transcript is moderately expressed in brain, abundant in thymus, and most abundant in white matter of the CNS.
